Macroscopic microscopy allows for the simultaneous recording of activity in various regions of the brain, giving the experimenter insight into how neuronal activity is coordinated during spontaneous and cognitive behaviors in mice. The use of widely available fluorescent activity indicators, such as calcium or voltage indicators, enables genetic coding, has a high signal-to-noise ratio and provides observation of the activity of specific cell types. This protocol is based on a combination of transgenic mouse technology and advanced recording techniques that allow the experimenter to image the entire mouse cortex with high spatial and temporal resolution. As neural activity indicators continue to be upgraded, new procedures are able to capture neural signals through the intact skull, reducing the potential for traumatic brain injury, inflammation, and tissue regeneration, which makes wide-field imaging ideally suited for longitudinal studies and high-throughput analyses.
